Morphological Changes and Distribution of Fibronectin in the Carrageenan-induced Arthritis

Abstract
|
|
|
The morphological changes and distribution of fibronectin were investigated in the carrageenan-induced arthritis of knee joints. The dose of 10mg crude carrageenan was injected twice into the knee joints via suprapatella pouch in adult rats and they were sacrificed at various intervals up to the seventh week. Histological examinations were done with H & E, alcian blue (pH 5.3), silver impregnation and Van Gieson stains. The distribution of fibronectin was observed by using Avidin-Biotin-Peroxidase complex(ABC) in immunoperoxidase technique. The results obtained were as follows.
1. The histological changes at early times disclosed an acute inflammatory reaction in the synovium and soft tissues such as adjacent ligament and tendon. And then, inflammatory cell infiltrates chiefly composed of carrageenan-laden macrophages and mononuclear cells were prominent. After the second week, with reduction of proteoglycan contents in the articular cartilage, granulation tissue was markedly formed in joint cavity, resulting in the prominent destruction of all articular cartilage.
2. Fibronectin appeared in acute inflammatory exudates and in chronic inflammatory reaction as fibrinous or fibrillar network. It showed significant increase in the young granulation tissue as dense extracellular matrix, with relative co distribution of reticular fibers. With maturation of granulation, it revealed a marked decrease in amount.
